PACITREX is a sterile, broad-spectrum third-generation cephalosporin antibiotic formulation by Pacitora Biotech containing Ceftriaxone 1 gm (I.P.) . It is indicated for the treatment of moderate to severe bacterial infections caused by susceptible organisms, including pneumonia, meningitis, sepsis, urinary tract infections (complicated) , intra-abdominal infections, skin and soft tissue infections, typhoid fever, bacterial endocarditis (as part of combination therapy), and uncomplicated gonorrhea.
Ceftriaxone works by inhibiting bacterial cell wall synthesis, leading to cell death (bactericidal). Its long elimination half-life (6-9 hours) allows for once-daily dosing, making it convenient for both inpatient and outpatient parenteral therapy. High tissue penetration ensures effective concentrations at sites of infection, including the cerebrospinal fluid (CSF) – making it a first-line agent for bacterial meningitis.

FAQs:
1. How is Pacitrex 1 gm administered?
For IM injection, reconstitute with 3.2 ml sterile water (or lidocaine 1% if approved by physician – lidocaine not supplied). For IV injection, reconstitute with 10 ml sterile water and inject slowly over 2-4 minutes. For IV infusion, dilute further in 50-100 ml compatible fluid.
2. Can Pacitrex be given to patients with penicillin allergy?
Cross-reactivity between penicillins and cephalosporins is approximately 5-10%. Use with caution only if absolutely necessary and have emergency equipment available. Do not use in patients with severe penicillin anaphylaxis.
3. Why is ceftriaxone not recommended for neonates with jaundice?
Ceftriaxone displaces bilirubin from albumin binding sites, increasing free bilirubin levels and risk of kernicterus (bilirubin-induced brain damage) in neonates.
4. Can Pacitrex be mixed with calcium-containing IV solutions?
In neonates (<28 days) – absolutely contraindicated (risk of fatal ceftriaxone-calcium precipitation). In older children and adults, ceftriaxone and IV calcium should not be administered simultaneously through the same IV line.
